The nervous system, hormonal axes, emotional context, blood vessels, medicines, sleep, stress and the relationship between partners all take part in it. That is why the phrase “peptides for libido” does not denote one class with identical action either.

PT-141, Kisspeptin-10 and oxytocin belong to different biological systems. Research into each of them starts from a different mechanism, and the quality and extent of the evidence are not equal.

PT-141, that is bremelanotide

PT-141 is the development designation for bremelanotide, a synthetic analogue linked to melanocortin receptors. Unlike substances that primarily affect blood flow, bremelanotide acts through central signalling pathways involved in sexual desire.

The American FDA approved bremelanotide in the form of a subcutaneous injection for a precisely defined group: premenopausal women with acquired, generalised hypoactive sexual desire disorder that causes significant distress and is not the consequence of another health condition, a medicine or a relationship problem. The approval does not cover men, postmenopausal women or “improvement of sexual performance”.

This regulatory fact is often passed on incorrectly. It shows that one particular formulation, dose and route of administration passed clinical assessment for one particular indication. It does not automatically confirm other routes of administration, other formulations or combinations.

In phase 3 trials, improvements in measures of sexual desire and a reduction in distress were recorded in some participants. At the same time, the official prescribing information lists important risks, including a transient rise in blood pressure, a drop in heart rate, nausea and the possibility of focal hyperpigmentation. That is another reason why the fact of approval must not be turned into a general recommendation.

Kisspeptin: the link between the reproductive axis and the brain’s processing of sexual signals

Kisspeptin is a natural signalling peptide with a key role in starting the reproductive hormonal axis. It activates the neurons that release gonadotropin-releasing hormone, thereby affecting the secretion of LH and FSH and the downstream function of the gonads.

More recent research has extended interest from reproductive hormones to emotional and sexual processing in the brain. In a randomised controlled trial in men with hypoactive sexual desire, intravenous administration of kisspeptin-54 changed the activity of networks linked to sexual processing and was associated with an increase in the physiological response to visual sexual stimuli.

That is an important proof of concept, but it should not be overstated. The trial was small, carried out in a single research centre and used an intravenous infusion of kisspeptin-54. The results cannot be transferred directly to Kisspeptin-10, to a sublingual format or to a fixed combination with other peptides.

Oxytocin: a hormone of social context with uneven results

Oxytocin is a peptide hormone known for its role in childbirth and breastfeeding, but it is also involved in social bonding, the processing of emotional signals and certain aspects of the sexual response. Its popular presentation as a simple “love hormone” is far too narrow.

Studies of intranasal oxytocin and sexual function have produced mixed results. In one smaller trial in couples, changes in orgasm intensity and satisfaction after intercourse were recorded, but not a consistent improvement in all subjective parameters. In a controlled trial in women, both oxytocin and placebo were associated with improvement over time, with no statistically significant difference between the treatments.

This shows how important expectations, context, the relationship with a partner and the method of measurement are in this field. Oxytocin’s biological involvement in the sexual response is not the same as confirmed efficacy of every external administration.

What does combining three different peptide directions mean?

In theory, a combination of PT-141, Kisspeptin-10 and oxytocin covers three levels:

melanocortin signalling, linked to central regulation of sexual desire;

the reproductive hormonal axis, in which kisspeptin has a key regulatory role;

social and emotional processing, in which oxytocin takes part.

The biological logic of a combination is not the same as clinical evidence. A fixed complex requires separate studies assessing stability, absorption, mutual interactions, safety and actual outcomes. The result of a study of one ingredient does not automatically add to the result of another.

A particularly important distinction: molecule, formulation and route of administration

When bremelanotide was approved as a subcutaneous injection, that does not confirm sublingual bremelanotide. When kisspeptin-54 was studied by intravenous infusion, that does not confirm Kisspeptin-10 administered through the oral mucosa. When oxytocin was studied intranasally, the result says nothing directly about the oromucosal format.

The route of administration determines how much of a substance reaches the circulation, how quickly, how long the exposure lasts and which side effects may occur. With peptides this difference is particularly large because of their sensitivity to enzymes and the limited permeability of membranes.

SORA Potency / Libido Complex

The SORA Potency / Libido Complex is presented for information as a set containing a vial with the combination of PT-141, Kisspeptin-10 and oxytocin, a 5 ml bottle with a buffered sublingual/oromucosal diluent and a 5 ml bottle with a dropper.

The composition of the set should not be interpreted as a proven therapy or as a substitute for a medical examination. A drop in libido can be linked to hormonal disorders, medicines, depression, cardiovascular problems, pain, stress and relationships, so assessing the cause takes precedence over random experimenting.

Conclusion

PT-141 has the strongest clinical documentation of these three ingredients, but only for a specific injectable formulation and a strictly defined population. Kisspeptin has promising early human data on sexual processing, while the results for oxytocin are contextual and uneven. A fixed sublingual combination of all three peptides requires data of its own.

Sources

FDA prescribing information for bremelanotide (Vyleesi)

Phase 3 trials of bremelanotide

Kisspeptin in men with hypoactive sexual desire

Kisspeptin and sexual processing in the brain

Controlled trial of intranasal oxytocin in women

An overview of the research on PT-141, kisspeptin and oxytocin in the context of sexual desire, with the clear limits of the existing clinical evidence.

[228

post-30969","post","type-post","status-publish","format-standard","has-post-thumbnail","hentry","category-peptides
["GET
